Biological Background: IRF5 Function and Localization
- IRF5 (Interferon regulatory factor 5) is a transcription factor that plays a critical role in innate immunity by activating expression of type I interferon (IFN-α/IFN-β) and inflammatory cytokines. Related references: PMID:11303025 PMID:15695821 PMID:22412986
- It mediates IFN induction downstream of endolysosomal toll-like receptors TLR7, TLR8, and TLR9 via the MyD88-dependent signaling pathway.
- IRF5 is a key transcription factor regulating the IFN response during SARS-CoV-2 infection. Related references: PMID:33440148
- The protein binds to interferon-stimulated response elements (ISRE) in the promoters of target genes and can efficiently activate both IFN-beta and IFN-alpha genes.

- The immunogen corresponds to the N-terminal region (aa 1-200) of human IRF5, which may result in detection of multiple isoforms if this region is conserved across splice variants.
- For Western blotting, a band at approximately 56 kDa is expected; consider using positive control lysates from human or rat cells treated with TLR agonists.
- For immunohistochemistry, validate antibody specificity by using appropriate tissue controls and considering the reported nuclear and cytoplasmic localization.
- In ELISA applications, this polyclonal antibody can be used as a capture or detection reagent; pairing with a monoclonal antibody may improve assay specificity.
