Biological Background: IRF3 Function and Localization

  • IRF3 is a key transcriptional regulator of type I interferon (IFN)-dependent immune responses, playing a critical role in innate immunity against DNA and RNA viruses. Related references: PMID:22394562, PMID:24049179, PMID:25636800
  • It regulates transcription of type I IFN genes (IFN-alpha and IFN-beta) and IFN-stimulated genes (ISG) by binding to an interferon-stimulated response element (ISRE) in their promoters. Related references: PMID:11846977, PMID:16846591, PMID:16979567
  • IRF3 is a more potent activator of the IFN-beta (IFNB) gene than the IFN-alpha (IFNA) gene and plays a critical role in both the early and late phases of the IFNA/B gene induction. Related references: PMID:16846591, PMID:16979567, PMID:20049431
  • Found in an inactive form in the cytoplasm of uninfected cells; following viral infection, double-stranded RNA (dsRNA), or toll-like receptor (TLR) signaling, it is phosphorylated by IKBKE and TBK1 kinases, inducing a conformational change that leads to dimerization and nuclear localization. Related references: PMID:22394562, PMID:25636800, PMID:27302953
  • In the nucleus, IRF3 associates with CREB binding protein (CREBBP) to form dsRNA-activated factor 1 (DRAF1), a complex that activates the transcription of type I IFN and ISG genes. Related references: PMID:16154084, PMID:27302953, PMID:33440148
  • In response to Sendai virus infection, IRF3 is recruited to mitochondrion and forms an apoptosis complex TOMM70:HSP90AA1:IRF3:BAX, inducing apoptosis. Related references: PMID:25609812
  • Expressed constitutively in a variety of tissues.
  • Post-translational modifications include phosphorylation (critical for activation), acetylation, and ubiquitination; alternative splicing generates additional isoforms.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 100–200 of human IRF3 (NP_001184051.1), a region within the N-terminal DNA-binding domain; consider using this region for epitope mapping or blocking experiments.
  • In Western blot, a band at approximately 47 kDa is expected; validate in relevant virus-infected or IFN-stimulated cell lines to confirm expression.
  • Cross-reactivity with mouse and rat IRF3 is reported; verify sequence homology in the immunogen region if using these species.
  • For ChIP applications, optimize cross-linking and sonication conditions due to IRF3's dynamic nucleocytoplasmic shuttling.
  • For IF/ICC, use appropriate fixation and permeabilization methods to detect both cytoplasmic and nuclear localization.
