Biological Background: IRE1 Function and Localization

IRE1 (ERN1) is a serine/threonine-protein kinase and endoribonuclease that serves as a critical sensor for the unfolded protein response (UPR) in the endoplasmic reticulum (ER). In unstressed cells, its luminal domain is bound by the chaperone HSPA5/BiP; upon accumulation of misfolded proteins, HSPA5/BiP dissociates, allowing IRE1 to homodimerize, autophosphorylate, and activate its endoribonuclease domain. - The immunogen corresponds to amino acids 286–435 in the cytoplasmic region of human IRE1, which includes part of the kinase domain. This region is well-conserved across human, mouse, and rat, consistent with the reported cross-reactivity.
- For Western blot (WB), a band at approximately 110 kDa is expected; use reducing conditions and validate with appropriate loading controls and ER stress inducers (e.g., tunicamycin, thapsigargin) to confirm specificity.
- In immunofluorescence (IF/ICC) and immunohistochemistry (IHC-P), the protein is localized to the endoplasmic reticulum; permeabilization is required, and co-staining with ER markers may aid validation.
