Biological Background: IRE1 Function and Localization

  • IRE1 (also known as IRE1P, IRE1a, or hIRE1p) is a ~110 kDa serine/threonine-protein kinase and endoribonuclease encoded by the ERN1 gene in humans.
  • IRE1 serves as a critical sensor in the endoplasmic reticulum (ER) unfolded protein response (UPR). Under basal conditions, its luminal domain is kept inactive by binding to HSPA5/BiP. Accumulation of misfolded proteins triggers release, homodimerization, autophosphorylation, and activation of its endoribonuclease domain. Related references: PMID:11175748, PMID:11779464, PMID:12637535, PMID:21317875
  • The activated endoribonuclease domain specifically cleaves 26 nucleotides from XBP1 mRNA, generating a spliced transcript that encodes a potent transcription factor driving UPR target gene expression. Related references: PMID:11779464, PMID:21317875, PMID:24508390
  • IRE1 is an ER-resident transmembrane protein, with both its kinase and RNase domains facing the cytoplasm, and it functions as a proximal signal for ER stress-mediated unconventional trafficking pathways, such as CFTR delivery to the plasma membrane via GORASP2 and SEC16A modulation. Related references: PMID:21884936, PMID:28067262
  • The protein undergoes multiple post-translational modifications including phosphorylation, ADP-ribosylation, and glycosylation, and harbors key functional motifs: an ATP-binding kinase domain, a disulfide bond-stabilized luminal sensor, and a metallo-dependent endoribonuclease active site.
  • IRE1 is ubiquitously expressed in human tissues, with particularly high levels in the pancreas, consistent with its central role in ER proteostasis.
  • Alternatively spliced isoforms and polymorphisms of ERN1 may fine-tune UPR signaling intensity and contribute to cell fate decisions between adaptation and apoptosis.

Experimental Guidance and Technical Tips

  • The immunogen used for this antibody is a recombinant fusion protein covering amino acids 286–435 of human IRE1 (NP_001424.3), which lies within the juxtamembrane and kinase domain regions. This region is conserved across human, mouse, and rat, supporting the reported cross-reactivity.
  • For Western blot applications, consider using ER stress inducers (e.g., tunicamycin, thapsigargin) to enhance IRE1 detection, and validate the antibody in the relevant sample system with appropriate loading controls.
  • In ELISA, optimize coating and detection conditions as the antibody recognizes a linear epitope within the immunogen sequence.
  • The predicted molecular weight is ~110 kDa, but post-translational modifications and activation-induced phosphorylation may cause slight mobility shifts; confirm with molecular weight markers.
