Biological Background: IRAK4 Function and Localization

  • Interleukin-1 receptor-associated kinase 4 (IRAK4), also known as renal carcinoma antigen NY-REN-64, is encoded by the IRAK4 gene in humans.
  • Serine/threonine-protein kinase that plays a critical role in initiating innate immune response against foreign pathogens; involved in Toll-like receptor (TLR) and IL-1R signaling pathways (PubMed:17878374). Related references: PMID:17878374
  • Upon TLR activation, IRAK4 is rapidly recruited by MYD88 to form the Myddosome complex together with IRAK2, where it phosphorylates and stimulates IRAK1 kinase activity.
  • Phosphorylates E3 ubiquitin ligases Pellino proteins (PELI1, PELI2, PELI3) to promote IRAK1 polyubiquitination, enabling IKBKG/NEMO binding and subsequent NF-kappa-B activation via the IKK complex.
  • Additionally, phosphorylates TIRAP to promote its ubiquitination and degradation, and regulates NADPH oxidase activation by phosphorylating NCF1.
  • Subcellular location is cytoplasmic.
  • Alternative splicing produces multiple isoforms; the protein is associated with disease variants such as immunodeficiency (IMD67) and contains kinase-essential motifs (ATP-binding, magnesium, nucleotide-binding).

  • KD-validated for Western blot, this antibody can be used to confirm knockdown efficiency or detect endogenous IRAK4 in human cell lysates.
  • For ELISA applications, direct or capture formats may be feasible; users should optimize coating conditions and validate in relevant sample systems.
