Biological Background: IPO7 Function and Localization
- Importin-7 (IPO7), also known as Ran-binding protein 7 (RanBP7), functions as a nuclear transport receptor that can act autonomously or as an adapter in association with importin-beta subunit KPNB1.
- Autonomously mediates the nuclear import of ribosomal proteins RPL23A, RPS7 and RPL5 via a Ran-dependent mechanism. Related references: PMID:11682607
- In association with KPNB1, facilitates the nuclear import of H1 histone, and in vitro can transport core histones H2A, H2B, H3 and H4.
- Plays a role in odontoblast differentiation by promoting nuclear translocation of transcription factors such as DLX3, KLF4 and SMAD2, and regulates MSX1 expression via SMAD1 import.
- Inhibits osteoblast differentiation by preventing RUNX2 nuclear translocation and subsequent target gene transcription.
- Subcellular localization is both cytoplasmic and nuclear, consistent with its function as a shuttling transport receptor.
- Post-translationally modified by acetylation and phosphorylation; identified by mass spectrometry in proteomic studies.
- During HIV-1 infection, Importin-7 mediates nuclear import of the viral reverse transcription complex integrase and Rev protein.

- The immunogen corresponds to a C-terminal peptide (aa 900–end) of human IPO7, which may confer reactivity with isoforms containing this region.
- For western blot, a band near 120 kDa is expected; consider using reducing conditions and appropriate loading controls.
- In immunofluorescence, signals should be observable in both nucleus and cytoplasm; include appropriate controls for nuclear transport studies.
- For ELISA, optimize coating antigen and antibody concentration empirically; the polyclonal nature may reduce sensitivity compared to monoclonal alternatives.
- Cross-reactivity with mouse and rat is indicated; validate in the relevant species sample system before quantitative assays.
