Biological Background: Importin-5 Function and Localization
- Importin-5 (also known as importin subunit beta-3, karyopherin beta-3, or RanBP5) is a key member of the importin beta family of nuclear transport receptors.
- Serves as a receptor for nuclear localization signals (NLS) in cargo substrates, mediating docking to the nuclear pore complex and energy-dependent, Ran-mediated translocation into the nucleus (By similarity).
- Mediates the nuclear import of ribosomal proteins RPL23A, RPS7, and RPL5, and also imports histones H2A, H2B, H3, and H4 in vitro.

PMID:11682607 PMID:9687515 - Plays a role in HIV-1 infection by binding and mediating the nuclear import of HIV-1 Rev, highlighting its involvement in host-virus interactions.
- Localized predominantly in the cytoplasm, nucleus, and nucleolus, consistent with its shuttling role.
- Undergoes post-translational modifications including acetylation and phosphorylation, which may regulate its activity.
- Alternative splicing produces multiple isoforms, contributing to functional diversity.
Experimental Guidance and Technical Tips
- The immunogen (aa 20–300) targets the N-terminal region of IPO5, which may be effective for detecting both full-length and certain truncated forms.
- For Western blotting (WB), anticipate a band near 124 kDa; use suitable loading controls and validate with known positive and negative samples.
- In ELISA applications, the recombinant protein used as immunogen can serve as a reference standard for quantification.
- Given cross-reactivity with mouse and rat, verify reactivity in these species with appropriate tissue or cell lysates.
- Always optimize antibody dilutions and experimental conditions for your specific assay system.
