Biological Background: IPA1 Function and Localization

  • SPL14/IPA1 is a SQUAMOSA promoter-binding-like transcription factor containing an SBP-box DNA-binding domain. It recognizes the 5'-GTAC-3' core motif and, via interaction with PCF1/2, can target the TCP motif 5'-TGGGCC/T-3'. Related references: PMID:24170127

  • As a master regulator of rice plant architecture, IPA1 suppresses tillering and promotes panicle branching, leading to ideal plant architecture. Related references: PMID:20495564, PMID:20495565, PMID:22960246

  • IPA1 significantly enhances grain yield by optimizing panicle structure and seed setting. Related references: PMID:20495564, PMID:20495565, PMID:26519999

  • Direct transcriptional targets include TB1 (tiller bud growth suppressor) and DEP1 (plant height and panicle length regulator); IPA1 binds and activates their expression. Related references: PMID:24170127

  • In strigolactone signaling, IPA1 acts downstream of D53 to mediate tiller number regulation and participates in a negative feedback loop controlling D53 expression. Related references: PMID:28809396

  • Following infection by Magnaporthe oryzae, IPA1 is phosphorylated at Ser-163, shifting its DNA-binding preference from the DEP1 promoter to the WRKY45 promoter, thereby activating pathogen defense genes. It also stabilizes SLR1 to reduce gibberellin-mediated susceptibility. Related references: PMID:30190406, PMID:30886331

  • IPA1 is localized in the nucleus and is highly expressed in young panicles, shoot apices, and branch primordia, consistent with its role in meristem development.

  • The protein contains conserved SBP-box and zinc-finger motifs, undergoes phosphorylation at Ser-163, and may be regulated by ubiquitination.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 209–417 of rice IPA1; verify antibody specificity for the full-length protein (~42 kDa) in Western blot using appropriate molecular weight markers.
  • For Western blot, total protein extracts from young rice panicles or shoot apices are recommended, as IPA1 expression is highest in these tissues.
  • Optimize antibody dilution for your specific sample type; a starting dilution of 1:500–1:2000 is typical for polyclonal antibodies in WB, but empirical determination is essential.
  • In ELISA, recombinantly expressed IPA1 protein can be used as a positive control to establish standard curves.
  • Cross-reactivity has been confirmed only with Oryza sativa; if working with other species, sequence homology should be checked before use.
  • This is a polyclonal antibody; lot-to-lot consistency should be monitored by including reference samples in each experiment.
