- INVS encodes Inversin, also known as nephrocystin-2, a multifunctional protein with ANK repeats and calmodulin-binding domains.
- Critical for normal renal development and establishment of the left-right body axis during embryogenesis.
- Acts as a molecular switch between canonical and non-canonical Wnt signaling pathways by targeting cytoplasmic DVL1 for ubiquitin-proteasome degradation, thereby inhibiting canonical Wnt signaling.
- Collaborates with nephrocystin proteins NPHP1, NPHP4, and RPGRIP1L/NPHP8 to organize apical junctions in kidney tubular epithelial cells.
- Localizes to multiple subcellular compartments including the cytoplasm, cytoskeleton, mitotic spindle, membrane, nucleus, and primary cilium.
- Widely expressed in tissues with strong enrichment at the primary cilia of renal tubular cells, consistent with its role in ciliopathy-associated renal disease.
- Post-translational modifications include phosphorylation and hydroxylation, and the protein is subject to alternative splicing and ubiquitin conjugation.
- Mutations in INVS are associated with nephronophthisis type 2, an autosomal recessive cystic kidney disease.
