  • Critical for normal renal development and establishment of the left-right body axis during embryogenesis.
  • Acts as a molecular switch between canonical and non-canonical Wnt signaling pathways by targeting cytoplasmic DVL1 for ubiquitin-proteasome degradation, thereby inhibiting canonical Wnt signaling.
  • Collaborates with nephrocystin proteins NPHP1, NPHP4, and RPGRIP1L/NPHP8 to organize apical junctions in kidney tubular epithelial cells.
  • Localizes to multiple subcellular compartments including the cytoplasm, cytoskeleton, mitotic spindle, membrane, nucleus, and primary cilium.
  • Widely expressed in tissues with strong enrichment at the primary cilia of renal tubular cells, consistent with its role in ciliopathy-associated renal disease.
