Biological Background: INTS8 Function and Localization
- INTS8 (Integrator complex subunit 8), also known as protein kaonashi-1, is a core component of the integrator complex, a multi-subunit assembly that terminates RNA polymerase II (Pol II) transcription at promoter-proximal regions. Related references: PMID:28542170 PMID:33243860 PMID:34004147
- INTS8 is specifically required for the recruitment of protein phosphatase 2A (PP2A) to the transcription pause-release checkpoint, a critical step in quality control during elongation. Related references: PMID:32966759 PMID:33243860 PMID:34004147
- The integrator complex terminates transcription via three coordinated enzymatic activities: CTD dephosphorylation of Pol II and SUPT5H, endonucleolytic cleavage of nascent RNA, and DNA release of the polymerase. Related references: PMID:33243860 PMID:34004147 PMID:38570683
- Beyond mRNA synthesis, INTS8-containing integrator also terminates transcription of non-coding Pol II products, including enhancer RNAs (eRNAs), small nuclear RNAs (snRNAs), telomerase RNAs, and long non-coding RNAs (lncRNAs). Related references: PMID:16239144
- INTS8 localizes to the nucleus and is found on chromosomes, consistent with its direct involvement in transcription regulation.
- The protein contains tetratricopeptide (TPR) repeat motifs, which are likely to mediate protein-protein interactions essential for integrator complex assembly.
- Alternative splicing generates multiple INTS8 isoforms, and the protein is subject to phosphorylation; pathogenic variants in INTS8 are associated with intellectual disability.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 600-800 of human INTS8, a region containing TPR repeats that may be accessible under denaturing conditions, supporting use in Western blotting (WB).
- For WB, consider using whole-cell lysates from human or mouse cell lines; validate antibody specificity with recombinant INTS8 or knockdown/knockout controls in your experimental system.
- This polyclonal antibody can be employed for ELISA-based detection of INTS8; pairing with a matched detection antibody may improve sensitivity and dynamic range.
- Optimize antibody dilution and incubation conditions for your specific sample type and application, as performance can vary with target abundance and matrix.
