Biological Background: INTS7 Function and Localization

  • INTS7 is a component of the integrator complex, which terminates RNA polymerase II (Pol II) transcription in the promoter-proximal region of genes. Related references: PMID:33243860, PMID:38570683
  • The integrator complex functions as a quality checkpoint during transcription elongation, driving premature termination of unfavorably configured transcripts. Related references: PMID:33243860, PMID:38570683
  • The complex terminates transcription by dephosphorylating the C-terminal domain (CTD) of Pol II and SUPT5H/SPT5, cleaving the nascent RNA via endonuclease activity, and promoting Pol II release from DNA. Related references: PMID:33243860, PMID:38570683
  • INTS7-containing integrator complex also terminates synthesis of non-coding Pol II transcripts, including enhancer RNAs (eRNAs), small nuclear RNAs (snRNAs), telomerase RNAs, and long non-coding RNAs (lncRNAs). Related references: PMID:16239144
  • INTS7 contributes to DNA damage response (DDR) signaling specifically during the S phase of the cell cycle. Related references: PMID:21659603
  • Localizes to the nucleus and chromosomes, with additional presence in the cytoplasm.
  • Subject to alternative splicing and post-translational phosphorylation, indicating regulatory diversity.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to a region near the C-terminus (aa 702-948) of human INTS7; thus, the antibody may detect full-length INTS7 as well as isoforms that include this segment.
  • For Western blot, consider using a denaturing gel and validating with appropriate positive and negative controls; the predicted molecular weight is ~107 kDa.
  • Cross-reactivity is reported for human and mouse; perform species-specific validation if using in other models.
  • For ELISA, titrate the antibody and pair with a suitable secondary antibody to optimize signal-to-noise ratio.
  • As a polyclonal antibody, lot-to-lot consistency should be verified for critical quantitative applications.
