Biological Background: INTS5 Function and Localization

  • INTS5 (Integrator complex subunit 5), also known as INT5 or KIAA1698, is a component of the integrator complex, a multiprotein assembly that regulates RNA polymerase II (Pol II) transcription termination.
  • The integrator complex terminates Pol II transcription in the promoter-proximal region by catalyzing dephosphorylation of the Pol II C-terminal domain and SUPT5H, endonucleolytic cleavage of nascent RNA, and promoting Pol II release from DNA. Related references: PMID:33243860 PMID:38570683
  • INTS5-containing integrator also terminates synthesis of non-coding Pol II transcripts, including enhancer RNAs (eRNAs), small nuclear RNAs (snRNAs), telomerase RNAs, and long non-coding RNAs (lncRNAs). Related references: PMID:16239144
  • The protein mediates recruitment of cytoplasmic dynein to the nuclear envelope, likely as part of the integrator complex. Related references: PMID:23904267
