Biological Background: Integrin-β1/CD29 Function and Localization

  • Integrin beta-1 (CD29) is a transmembrane receptor subunit that non-covalently pairs with various alpha subunits to form functional integrin heterodimers. These complexes act as receptors for extracellular matrix proteins such as collagen, fibronectin, laminin, and vitronectin.
  • The alpha/beta-1 integrins mediate cell adhesion, spreading, migration, and matrix remodeling. They are essential for processes like angiogenesis, myogenesis, osteoblast function, and cytokinesis.
  • ITGA4:ITGB1 and ITGA5:ITGB1 have specialized ligand interactions, including fractalkine (CX3CL1) signaling, PLA2G2A-enhanced ligand binding, fibrillin-1 adhesion, and IL1B/CD40LG signaling. Related references: PMID:23125415, PMID:24789099, PMID:18635536, PMID:25398877, PMID:12807887, PMID:17158881, PMID:33962943, PMID:29030430, PMID:31331973, PMID:35802072
  • Integrin beta-1 is predominantly localized to the plasma membrane, concentrating at focal adhesions, lamellipodia, invadopodia, and the cleavage furrow. It also cycles through recycling endosomes and is found in melanosomes and the sarcolemma.
  • ITGB1 is widely expressed in many tissues, with detection in placenta, vascular smooth muscle, skin, liver, muscle, and hematopoietic cells. Isoform 5 is restricted to striated muscle (skeletal and cardiac). Related references: PMID:33962943, PMID:1551917, PMID:7545396, PMID:7681433
  • Integrin beta-1 serves as a receptor for multiple pathogens, including viruses (echovirus, CMV, EBV, parvovirus B19, rotavirus, reovirus, HIV-1) and fungi of the order Mucorales, where ITGA3:ITGB1 binds fungal CotH proteins to mediate epithelial invasion. Related references: PMID:32487760
  • The protein undergoes various post-translational modifications such as acetylation, phosphorylation, and ubiquitination, and its integrin function depends on divalent cations (Ca²⁺, Mg²⁺). It contains an EGF-like domain and multiple disulfide bonds.

Experimental Guidance and Technical Tips

  • For Western blot, the predicted molecular weight is 88 kDa, but the native protein often migrates at 130–140 kDa under non-reducing conditions due to glycosylation and disulfide bonds; consider using reducing and non-reducing conditions to confirm specificity.
  • In immunohistochemistry (IHC-P), strong membrane staining is expected; include appropriate antigen retrieval steps (e.g., citrate buffer, pH 6.0) and a validation control.
  • For immunofluorescence/ICC, this antibody may label plasma membrane and focal adhesions; co-staining with focal adhesion markers (e.g., paxillin) can confirm specificity.
  • ELISA performance may depend on coating antigen and detection system; cross-reactivity with mouse provides utility for murine samples.
