Biological Background: Integrin-β1/CD29 Function and Localization

  • Integrin beta-1 (ITGB1, CD29) is a transmembrane receptor that pairs with various alpha subunits to form functional integrin heterodimers. It binds to extracellular matrix proteins including collagen, fibronectin, laminin, and vitronectin, mediating cell adhesion and signaling.
  • The beta-1 integrin subfamily includes receptors for collagen, fibronectin, fibrinogen, laminin, VCAM1, and other ligands, playing key roles in processes like angiogenesis, immune cell trafficking, and tissue development.
  • ITGA4:ITGB1 binds to fractalkine (CX3CL1) and may act as its coreceptor in CX3CR1-dependent signaling, while ITGA5:ITGB1 acts as a receptor for IL1B and soluble CD40LG, contributing to inflammation and immune responses. Related references:
    PMID:23125415, PMID:24789099, PMID:29030430, PMID:31331973
  • Subcellularly, ITGB1 localizes to the cell membrane and is enriched at focal adhesions, invadopodia, ruffles, and lamellipodia, consistent with its role in cell migration and invasion.
  • ITGB1 is widely expressed, with protein detected in placenta, vascular smooth muscle, skin, liver, skeletal muscle, cardiac muscle, and hematopoietic cells. Isoform 5 is specifically expressed in striated muscles.
  • Post-translational modifications include phosphorylation, acetylation, and ubiquitin conjugation. The protein contains EGF-like domains and requires calcium and magnesium for ligand binding.
  • ITGB1 serves as a receptor for mucormycosis-causing fungi (order Mucorales) by interacting with CotH proteins, facilitating epithelial invasion. Related references:
    PMID:32487760
  • Beyond physiological functions, ITGB1 is exploited as a host cell receptor by numerous pathogens, including human echoviruses, rotavirus, reovirus, and herpesviruses, and facilitates HIV-1 Tat-induced Kaposi's sarcoma angiogenesis.

Experimental Guidance and Technical Tips

  • The immunogen spans a large extracellular portion (aa 1-728) of the protein, making it suitable for detecting both full-length and certain truncated isoforms.
  • For Western blot, a band around 88 kDa is expected; consider using appropriate molecular weight markers and loading controls.
  • Due to cross-reactivity with mouse and rat, this antibody may be useful for studies in these species, but validation in the relevant sample system is recommended.
  • In immunofluorescence, the antibody can be used to visualize integrin β1 localization at focal adhesions and lamellipodia; optimize fixation and permeabilization conditions for your cell type.
  • For flow cytometry, test the antibody on live or fixed cells to assess surface expression of CD29; include appropriate isotype controls.
