Biological Background: Integrin beta-3 (ITGB3/CD61) Function and Localization
- Integrin beta-3 (ITGB3), also known as CD61 or GPIIIa, is a transmembrane receptor encoded by the ITGB3 gene. Two major isoforms exist: beta-3A, widely expressed, and beta-3C, specifically in prostate and testis.
- In platelets, ITGB3 pairs with ITGA2B to form the fibrinogen receptor alpha-IIb/beta-3, which upon activation mediates platelet aggregation essential for hemostasis. Related references: PMID:9111081
- As part of integrin alpha-V/beta-3, ITGB3 binds RGD-containing extracellular matrix ligands and various growth factors (FGF1, FGF2, IGF1, IGF2, IL1B) to regulate cell adhesion, migration, proliferation, and survival. It also serves as a coreceptor for CX3CL1 and NRG1 signaling. Related references: PMID:18441324 PMID:28302677 PMID:19578119 PMID:28873464 PMID:29030430 PMID:23125415 PMID:24789099 PMID:20682778
- It localizes at the cell membrane, enriched in focal adhesions, lamellipodia, and postsynaptic membranes.
- ITGB3 is a glycoprotein with multiple EGF-like domains, disulfide bonds, phosphorylation sites, and requires calcium and magnesium for function.
- ITGB3 acts as a host cell receptor for several viruses, including Herpes virus 8, Coxsackievirus A9, Hantaan virus, Cytomegalovirus, Human metapneumovirus, and West Nile virus.
- In the brain, ITGB3 participates in synaptic transmission and plasticity, regulating serotonin reuptake and AMPA receptor trafficking (By similarity).
Experimental Guidance and Technical Tips
- The immunogen covers the C-terminal region (aa 609-788) of human ITGB3, including part of the extracellular domain and the full transmembrane/cytoplasmic regions; thus, the antibody likely detects total ITGB3 and may recognize precursor or cleaved forms under appropriate conditions.
- For Western blot, the predicted molecular weight is ~87 kDa under reducing conditions. Validate in platelet lysates (for alpha-IIb/beta-3) or endothelial cells (for alpha-V/beta-3) from human and mouse. Non‑reducing conditions may be considered to preserve disulfide-linked epitopes.
- In ELISA applications, this polyclonal antibody can be utilized as a capture or detection reagent for quantifying ITGB3 levels. Careful titration and pairing with a monoclonal detection antibody are recommended.
- As a rabbit polyclonal, lot-to-lot performance may vary; always confirm specificity in the relevant sample system before quantitative assays.
