Biological Background: Integrin alpha-6/ITGA6 Function and Localization
- Integrin alpha-6 (ITGA6), also designated CD49f or VLA-6, is a member of the integrin alpha chain family that heterodimerizes with beta-1 or beta-4 subunits to form laminin receptors.
- The ITGA6:ITGB1 complex functions as a laminin receptor on platelets and is present in oocytes, where it is implicated in sperm-egg fusion (By similarity).
- In epithelial cells, ITGA6:ITGB4 serves as a laminin receptor and is a key structural component of hemidesmosomes (By similarity).
- ITGA6:ITGB4 binds to neuregulin-1 (NRG1) via its EGF domain, an interaction essential for NRG1-ERBB signaling. Related references: PMID:20682778
- ITGA6:ITGB4 binds to insulin-like growth factors IGF1 and IGF2, interactions that are essential for IGF1 and IGF2 signaling, respectively. Related references: PMID:22351760 PMID:28873464
- The protein undergoes proteolytic cleavage on a pair of basic residues to yield the mature alpha-6 heavy and light chains; it is also palmitoylated and contains multiple disulfide bonds and calcium-binding sites.
- Multiple isoforms are generated by alternative splicing, with the ubiquitously expressed isoform containing the X1 cytoplasmic segment and a longer X1X2 form found in heart, kidney, placenta, and other tissues.
- Processed ITGA6 is expressed at low levels in normal prostate tissue and is upregulated in prostate cancer.
- ITGA6 localizes to the cell membrane as a single-pass type I membrane protein.
Experimental Guidance and Technical Tips
- The immunogen sequence corresponds to an extracellular region (amino acids 505–663), making this antibody suitable for detecting cell-surface or native ITGA6 in non‑denaturing assays such as immunofluorescence.
- For western blotting, expect a major band at ~127 kDa; always include appropriate molecular‑weight markers and validate in your sample matrix due to potential glycosylation heterogeneity.
- In immunofluorescence and immunohistochemistry, staining should be membrane‑associated; co‑staining with a membrane marker is recommended to confirm specificity.
- ELISA performance may benefit from using recombinant integrin alpha‑6 protein or cell lysates as coating antigens; titrate antibody concentration for optimal signal‑to‑noise ratio.
