Biological Background: Integrin alpha-6 (ITGA6/CD49f) Function and Localization
- Integrin alpha-6 (ITGA6/CD49f) is a cell surface receptor that forms heterodimers with integrin beta-1 (ITGB1) or beta-4 (ITGB4) to mediate cell adhesion to laminin, playing critical roles in platelet function, epithelial integrity, and sperm-egg fusion.
- The ITGA6:ITGB4 complex acts as a receptor for neuregulin-1 (NRG1), and this interaction is essential for NRG1-ERBB signaling. Related references: PMID:20682778
- ITGA6:ITGB4 also binds insulin-like growth factor 1 (IGF1) and insulin-like growth factor 2 (IGF2), with these interactions being critical for IGF1 signaling and IGF2 signaling, respectively. Related references: PMID:22351760, PMID:28873464
- Integrin alpha-6 is localized to the cell membrane and is a key component of hemidesmosomes in epithelial cells, where it provides structural attachment to the basement membrane.
- This protein undergoes extensive post-translational modifications, including glycosylation, phosphorylation, palmitoylation, and proteolytic cleavage, which regulate its function and trafficking.
- Multiple isoforms of ITGA6 exist due to alternative splicing; tissue-specific expression patterns have been reported, and processed forms of the protein are upregulated in prostate cancer tissues. Related references: PMID:15023541
Experimental Guidance and Technical Tips
- The immunogen corresponds to a region within the extracellular domain of human ITGA6, which may be conserved across species, consistent with reported cross-reactivity to mouse and rat.
- For Western blotting, the predicted molecular weight is approximately 127 kDa; consider optimizing electrophoresis and transfer conditions for large proteins.
- In ELISA, pairing with an appropriate detection antibody for capture assays may be evaluated.
- Validate the antibody in your specific sample system and application conditions to ensure optimal performance.
