Biological Background: ITGA5/CD49e Function and Localization
- Integrin alpha-5 (ITGA5) is a type I transmembrane protein, also known as CD49e, VLA-5 alpha chain, and fibronectin receptor subunit alpha.
- ITGA5 pairs with integrin beta-1 (ITGB1) to form the fibronectin receptor, which binds to the RGD sequence in fibronectin and fibrinogen, mediating cell adhesion and signaling (PubMed:18635536, PubMed:25398877, PubMed:33962943).

PMID:18635536 PMID:25398877 PMID:33962943 - ITGA5:ITGB1 also acts as a receptor for fibrillin-1 (FBN1) and mediates RGD-dependent cell adhesion to FBN1 (PubMed:12807887, PubMed:17158881).
Related references:
PMID:12807887 PMID:17158881 - ITGA5:ITGB1 is involved in cytokine signaling, serving as a receptor for IL1B (via ITGA5:ITGB1) and soluble CD40LG (via ITGA5:ITGB3) (PubMed:29030430, PubMed:31331973).
Related references:
PMID:29030430 PMID:31331973 - In pathogen-host interactions, ITGA5:ITGB1 functions as a receptor for human metapneumovirus and HIV-1 Tat protein, facilitating viral entry or angiogenesis.
- Subcellularly, ITGA5 localizes to the cell membrane and cell junctions, particularly focal adhesions, where it connects the extracellular matrix to the actin cytoskeleton.
- Post-translational modifications: ITGA5 undergoes cleavage on pair of basic residues to yield heavy and light chains, and is N-glycosylated and phosphorylated, which may regulate its activity.
- According to UniProt, ITGA5 is expressed in placenta at the protein level, but may be present in other tissues as well.
Experimental Guidance and Technical Tips
- The immunogen used spans amino acids 42-995, encompassing nearly the entire extracellular domain. This antibody is likely to recognize the native conformation, making it suitable for detection of cell-surface ITGA5 in flow cytometry.
- The antibody is directly conjugated to , a bright fluorophore, eliminating the need for secondary staining and reducing background. Optimize the antibody concentration by titration in the target cell line or sample of interest.
- Validate the reactivity in both human and mouse systems, as cross-reactivity is indicated. Include appropriate positive and negative controls (e.g., known integrin α5-expressing cells and isotype control) in your FC panel.
- For fixation and permeabilization steps, verify if the antibody recognizes the denatured epitope; if not, surface staining is recommended.
- As integrin α5 is involved in adhesion and signaling, consider using this antibody in conjunction with functional assays to correlate surface expression with activity.
