Biological Background: Integrin alpha-4 Function and Localization
- Integrin alpha-4 (ITGA4, CD49d) forms heterodimers with integrin beta-1 (ITGB1) to form VLA-4, and with integrin beta-7 to form LPAM-1, serving as receptors for fibronectin, VCAM1, and MADCAM1.
- Through VLA-4, ITGA4 recognizes the Q-I-D-S sequence in VCAM1, while alpha-4/beta-7 recognizes the L-D-T sequence in MADCAM1, mediating leukocyte rolling, firm adhesion, and transmigration.
- ITGA4:ITGB1 binds to fractalkine (CX3CL1) and may act as its coreceptor in CX3CR1-dependent signaling. Related references: PMID:23125415
