Biological Background: Integrin alpha-3 Function and Localization
- Integrin alpha-3, also known as CD49c, FRP-2, GAPB3, or VLA-3 subunit alpha, is a member of the integrin alpha chain family.
- It combines with integrin beta-1 to form the VLA-3 receptor, which binds to extracellular matrix proteins including fibronectin, laminin, collagen, epiligrin, thrombospondin, and CSPG4.
- This integrin heterodimer mediates cell adhesion, invadopodia formation, and matrix degradation, promoting cell invasion. It also facilitates endothelial cell migration via interaction with LGALS3 and CSPG4.
- In the context of microbial infection, ITGA3:ITGB1 may serve as a receptor for Rhizopus delemar CotH7 in alveolar epithelial cells, potentially involved in pulmonary mucormycosis.
- Subcellularly, integrin alpha-3 localizes to the cell membrane, as well as to specialized membrane structures such as invadopodia and filopodia.
- The protein undergoes proteolytic cleavage on basic residues, and is subject to post-translational modifications including glycosylation, palmitoylation, and phosphorylation.
- Two isoforms exist due to alternative splicing: isoform 1 is widely expressed, while isoform 2 is expressed in brain and heart, with distinct vascular distribution patterns.
- Keywords associated with ITGA3 include calcium, cell adhesion, cell membrane, integrin, receptor, and disease variants linked to epidermolysis bullosa.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 466-620 of human Integrin alpha 3, a region within the extracellular domain. This design may enable detection of total integrin alpha-3 protein.
- For IHC-P, consider optimizing antigen retrieval conditions and validate with known positive control tissues expressing ITGA3.
- For flow cytometry, confirm reactivity on cell lines with confirmed ITGA3 surface expression, and include appropriate isotype controls.
- For ELISA, evaluate pairing with a suitable capture antibody and assess sensitivity using recombinant integrin alpha-3 or cell lysates as antigen.
- As a rabbit monoclonal antibody, it typically offers high specificity and lot-to-lot consistency; however, user validation in the intended assay system is recommended.
