Biological Background: Insulin Receptor Function and Localization
- Insulin receptor (IR) is a receptor tyrosine kinase that mediates the pleiotropic actions of insulin, including regulation of glucose uptake, cell growth, and differentiation.
- Upon insulin binding, IR phosphorylates insulin receptor substrates (IRS1-4) and SHC, activating the PI3K-AKT pathway for metabolic effects and the Ras-MAPK pathway for gene expression and cell growth.

- The receptor localizes to the cell membrane and, upon activation, internalizes to late endosomes and lysosomes.
- Post-translational modifications include autophosphorylation, glycosylation, and disulfide bond formation; it is a glycoprotein with tyrosine kinase activity.
- Expressed primarily in insulin-responsive tissues (liver, adipose, skeletal muscle) but also in many other tissues; overexpression is observed in various tumors.
- Mutations in INSR are associated with diabetes mellitus and severe insulin resistance syndromes.

- For Western blot, consider that the insulin receptor is processed into α (~135 kDa) and β (~95 kDa) subunits, leading to multiple bands depending on reducing conditions and sample preparation.
- Validate the antibody’s reactivity in your specific sample type (e.g., cell lysates, tissue extracts) using appropriate positive and negative controls, as the predicted molecular weight of the unprocessed precursor is ~156 kDa.
- For ELISA, titrate the antibody against recombinant insulin receptor or native receptor to optimize working concentrations, and include cross-reactivity validation for mouse and rat samples as indicated.
- Since the immunogen is a recombinant fragment (aa 475-570), the antibody may recognize both precursor and processed forms; confirm specificity via peptide competition or knockdown/knockout models if needed.
