Biological Background: INPP5K Function and Localization
- Encodes inositol polyphosphate 5-phosphatase K (INPP5K), also known as SKIP (skeletal muscle and kidney-enriched inositol phosphatase), which hydrolyzes the 5-phosphate from inositol 1,4,5-trisphosphate (IP3), inositol 1,3,4,5-tetrakisphosphate (IP4), phosphatidylinositol 4,5-bisphosphate (PIP2), and phosphatidylinositol 3,4,5-trisphosphate (PIP3). Related references: PMID:10753883, PMID:16824732
- Exhibits 6-fold higher affinity for PIP2 over IP3, positioning it as a key modulator of phosphoinositide signaling.
- Negatively regulates the assembly of the actin cytoskeleton, implicating INPP5K in cellular morphology and motility.
- Specifically controls insulin-dependent glucose uptake among inositol 3,4,5-trisphosphate phosphatases, serving as a critical regulator of insulin signaling in skeletal muscle (by similarity).
- Localizes predominantly to the endoplasmic reticulum and cytoplasm.
- Ubiquitously expressed with highest levels in skeletal muscle, heart, and kidney; accordingly, INPP5K is associated with tissue-specific functions and disorders.
- Keywords: Alternative splicing, disease variants linked to cataract, congenital muscular dystrophy, and intellectual disability; hydrolase, magnesium/metal-binding, lipid metabolism.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 149-448 of human INPP5K, a region conserved across the catalytic domain; consider verifying reactivity against mouse and rat orthologs to confirm cross-species specificity.
- For Western blot (WB), a band near 51 kDa is expected; however, alternative splicing and post-translational modifications may yield additional bands, so validate in your sample system.
- IF/ICC: Sample fixation and permeabilization conditions should be optimized, as INPP5K localizes to both cytoplasm and endoplasmic reticulum.
- ELISA: Suitable for detecting and quantifying INPP5K in cell lysates or purified protein; standard optimization of coating and blocking steps is advised.
