Biological Background: INPP5E Function and Localization

  • INPP5E, also known as phosphatidylinositol polyphosphate 5-phosphatase type IV, 72 kDa inositol polyphosphate 5-phosphatase, or pharbin, is encoded by the INPP5E gene (GeneID: 56623).
  • It specifically hydrolyzes the 5-phosphate from phosphatidylinositol-3,4,5-trisphosphate (PtdIns(3,4,5)P3), phosphatidylinositol 4,5-bisphosphate (PtdIns(4,5)P2), and phosphatidylinositol 3,5-bisphosphate (PtdIns(3,5)P2), being selective for lipid substrates and inactive toward water-soluble inositol phosphates. Related references: PMID:10764818
  • INPP5E can also remove the 5-phosphate from 1D-myo-inositol 1,4,5-trisphosphate (Ins(1,4,5)P3). Related references: PMID:40969890
  • The protein plays a critical role in primary cilium function by regulating ciliary growth and phosphoinositide 3-kinase (PI3K) signaling and stability.
  • Subcellular localization includes the cytoplasm, cytoskeleton, cilium axoneme, Golgi apparatus, cell membrane, ruffles, and nucleus.
  • It is expressed in a tissue‑specific manner, with detection in brain, heart, pancreas, testis, and spleen.
  • Mutations in INPP5E are associated with ciliopathies such as Joubert syndrome, and the protein is classified under keywords encompassing disease variant, intellectual disability, and obesity.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the C‑terminal region (aa 510‑630) of human INPP5E; thus, the antibody may recognize all isoforms sharing this domain. Verify isoform-specific expression in your target samples.
  • For Western blot, the expected molecular weight is approximately 70 kDa. Use appropriate loading controls and consider potential post‑translational modifications that may shift mobility.
  • In immunofluorescence (IF/ICC), localization to the primary cilium and Golgi apparatus may require optimized fixation/permeabilization protocols. Co‑staining with cilium markers (e.g., acetylated tubulin) is recommended.
  • This antibody is polyclonal; batch‑to‑batch variability is inherent. Validate specificity in knockout or knockdown controls where possible.
  • When using ELISA, include suitable negative controls to assess background signal, and optimize coating antigen concentration.
