Biological Background: INPP4A Function and Localization
-
INPP4A encodes type I inositol polyphosphate-4-phosphatase, which catalyzes the hydrolysis of the 4-position phosphate of phosphatidylinositol 3,4-bisphosphate (PtdIns(3,4)P2), as well as inositol 1,3,4-trisphosphate and inositol 1,4-bisphosphate.

By dephosphorylating phosphoinositides, INPP4A antagonizes the PI3K-AKT/PKB signaling pathway, thereby modulating cell cycle progression and cell survival.

The protein may protect neurons from excitotoxic cell death by regulating the synaptic localization of cell surface N-methyl-D-aspartate-type glutamate receptors (NMDARs) and NMDAR-mediated excitatory postsynaptic currents.
-
INPP4A localizes to multiple subcellular compartments, including early endosome and recycling endosome membranes, the cell membrane, nucleus, cytoplasm, and postsynaptic density.
-
Alternative splicing generates two isoforms: isoform 1 is expressed in platelets, MEG-01 megakaryocytes, and Jurkat T-cells, while isoform 2 is predominantly expressed in the brain.
-
The gene is associated with intellectual disability, as indicated by the disease variant keyword, highlighting its importance in neurodevelopment.
-
Other keywords include hydrolase, lipid metabolism, phosphoprotein, and proteomics identification.
Experimental Guidance and Technical Tips
- The immunogen (aa 1–250 of human INPP4A) is conserved across human, mouse, and rat, supporting the listed cross-reactivity; validate detection in your species of interest.
- For Western blotting, expect a band near 110 kDa in lysates from tissues or cells expressing INPP4A; use appropriate positive (e.g., brain lysate) and negative controls.
- In ELISA, the antibody may be used to capture or detect recombinant or endogenous INPP4A; optimize coating concentration and secondary detection conditions.
- Pre-incubation with the immunogen can serve as a specificity control to confirm band identity.
