Biological Background: iNOS Function and Localization
- Nitric oxide synthase, inducible (iNOS), also known as NOS2A, HEP-NOS, or NOS type II, is a calcium/calmodulin-dependent enzyme that catalyzes the production of nitric oxide (NO) from L-arginine, with diverse signaling functions. Related references: PMID:7504305, PMID:7531687, PMID:7544004
- In macrophages, iNOS-derived NO mediates tumoricidal and bactericidal actions, and the enzyme also exhibits nitrosylase activity, catalyzing cysteine S-nitrosylation of cytoplasmic targets like PTGS2/COX2. Related references: PMID:25417112

- For Western blot, a band around 131 kDa is expected; positive controls can include lysates from IFN-γ/LPS-stimulated RAW 264.7 (mouse) or THP-1 (human) macrophage cells.
- Cross-reactivity with mouse iNOS is reported; for other species, test in appropriate models before routine use.
- In ELISA, the antibody can be used for detection or capture; optimization of coating concentration and blocking conditions is recommended for best signal-to-noise ratio.
