Biological Background: iNOS Function and Localization
- Produces nitric oxide (NO), a messenger molecule with diverse functions. In macrophages, NO mediates tumoricidal and bactericidal actions. Also possesses nitrosylase activity, catalyzing cysteine S-nitrosylation of cytoplasmic proteins such as PTGS2/COX2. Related references: PMID:7503239, PMID:16373578
- As component of the iNOS-S100A8/9 transnitrosylase complex, is involved in the selective inflammatory stimulus-dependent S-nitrosylation of GAPDH, regulating the GAIT complex activity, and potentially modifies ANXA5, EZR, MSN, and VIM (By similarity).
- Involved in inflammation, enhancing the synthesis of pro-inflammatory mediators such as IL6 and IL8 (By similarity).
- Localizes predominantly to the cytoplasm and cytosol.
- Expressed mainly in macrophages.

- The immunogen corresponds to the N-terminal region (amino acids 1-200) of mouse iNOS, which may be responsible for the observed cross-reactivity with human and rat iNOS.
- For Western blot, expect a band at approximately 131 kDa; optimize gel percentage and transfer conditions accordingly.
- In immunofluorescence/ICC, validate fixation and permeabilization protocols as iNOS is a cytosolic protein.
- For ELISA, consider using the recombinant iNOS protein as a standard or capture reagent.
- Cross-reactivity with human and rat is reported; confirm sequence homology for species-specific applications.
