Biological Background: iNOS Function and Localization

  • Produces nitric oxide (NO), a messenger molecule with diverse functions. In macrophages, NO mediates tumoricidal and bactericidal actions. Also possesses nitrosylase activity, catalyzing cysteine S-nitrosylation of cytoplasmic proteins such as PTGS2/COX2. Related references: PMID:7503239, PMID:16373578
  • As component of the iNOS-S100A8/9 transnitrosylase complex, is involved in the selective inflammatory stimulus-dependent S-nitrosylation of GAPDH, regulating the GAIT complex activity, and potentially modifies ANXA5, EZR, MSN, and VIM (By similarity).
  • Involved in inflammation, enhancing the synthesis of pro-inflammatory mediators such as IL6 and IL8 (By similarity).
  • Localizes predominantly to the cytoplasm and cytosol.
  • Expressed mainly in macrophages.

  • In immunofluorescence/ICC, validate fixation and permeabilization protocols as iNOS is a cytosolic protein.
