Biological Background: Inhibin beta A Function and Localization
- Inhibin beta A (INHBA), also known as Activin beta-A chain or Erythroid differentiation protein (EDF), is a secreted protein that forms homodimers (Activin A) or heterodimers (Inhibin A with alpha chain/INHA) to regulate diverse physiological processes.
- Activin A signals through type I (ACVR1B, ACVR1C) and type II receptors (ACVR2A, ACVR2B, BMPR2), phosphorylating SMAD2/3 to form a complex with SMAD4 that regulates gene expression (PubMed:10652306, PubMed:24018044). It also activates non-canonical pathways like p38 MAPK, ERK1/2, and JNK (PubMed:16440334). Related references: PMID:10652306, PMID:24018044, PMID:16440334
- Involved in embryonic development, stem cell maintenance and differentiation, haematopoiesis, cell proliferation, and tissue fibrosis (PubMed:3194407, PubMed:16440334). Related references: PMID:3194407, PMID:16440334
- Inhibin A, the INHBA/INHA dimer, functions as a feedback regulator in the hypothalamic-pituitary-gonadal axis, inhibiting FSH secretion by antagonizing Activin A through betaglycan-mediated receptor sequestration.
- Promotes osteoblastic differentiation via the ACVRL1-SMAD1/5/9 pathway (PubMed:34948289) and can form non-signaling complexes with ACVR1 to inhibit BMP signaling (PubMed:26333933). Related references: PMID:34948289, PMID:26333933
- Subcellular localization: Secreted.
- Key keywords: Cleavage on pair of basic residues, disulfide bond, glycoprotein, growth factor, hormone, signal peptide, and proteomics identification.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-100) of human INHBA; expected band size in Western blot is ~47 kDa (pro-form). Validate in relevant cell or tissue lysates and consider potential detection of processed forms.
- For ELISA, the synthetic peptide immunogen provides suitability; use appropriate peptide or protein controls.
- The antibody cross-reacts with mouse, enabling studies in mouse models; validate its performance in your specific system.
