Biological Background: Inhibin alpha (INHA) Function and Localization
- Inhibin alpha chain (INHA) is the alpha subunit of the dimeric glycoprotein hormones inhibin A (alpha/beta-A) and inhibin B (alpha/beta-B), members of the transforming growth factor-beta (TGF-beta) superfamily.
- Inhibin A acts as a key regulator in the hypothalamic-pituitary-gonadal (HPG) axis, inhibiting the secretion of follicle-stimulating hormone (FSH) from anterior pituitary gonadotrope cells by antagonizing activin signaling; this involves binding to betaglycan and type II activin receptors.
- Inhibin B functions as a more potent suppressor of FSH release compared to inhibin A, operating as a competitive receptor antagonist that binds activin type II receptors with high affinity in the presence of the TGF-beta type III coreceptor (TGFBR3L). Related references: PMID:34910520
- INHA is secreted and widely expressed in reproductive tissues (ovarian granulosa cells, testicular Sertoli cells) as well as brain, placenta, adrenal cortex, and prostate, with circulating inhibin B predominantly in males and both inhibin A and B in females.
- Post-translational processing of INHA involves cleavage at paired basic residues to generate mature bioactive forms; it is glycosylated and contains multiple disulfide bonds, consistent with its secretory nature.
- Beyond reproductive axis regulation, inhibins/activins have diverse roles in germ cell development, erythroid differentiation, insulin secretion, nerve cell survival, embryonic development, and bone growth, underscoring their growth factor and hormone functions.
Experimental Guidance and Technical Tips
- For Western blotting (WB), a band corresponding to the mature inhibin alpha chain may be detected around 40 kDa; consider optimizing antibody dilution and verifying specificity using relevant positive controls or knockdown experiments.
- In immunohistochemistry (IHC-P), antigen retrieval methods (heat-mediated or enzymatic) may be required to expose the epitope; validate staining in tissue sections known to express INHA, such as ovary or testis.
- For ELISA applications, this polyclonal antibody can be used as a capture or detection reagent; pairing with an appropriate monoclonal antibody is recommended for sandwich assays; cross-reactivity to mouse and rat extends its utility in animal models.
- Given the synthetic peptide immunogen spanning residues 200‑300 of human INHA, confirm that the antibody recognizes both precursor and mature forms under non-reducing/reducing conditions if relevant.
