Biological Background: Importin Alpha 5 Function and Localization
- Importin alpha-5 (KPNA1) is a member of the importin alpha family of nuclear transport receptors and is also referred to as karyopherin subunit alpha-1, nucleoprotein interactor 1 (NPI-1), RAG cohort protein 2, and SRP1-beta.
- It serves as an adapter that recognizes classical nuclear localization signals (NLS) on cargo proteins and docks them to importin beta (KPNB1) for transport through the nuclear pore complex, a process essential for many cellular functions. Related references: PMID:27713473, PMID:7892216, PMID:8692858
- The import complex is translocated via a Ran-GTP-dependent mechanism, with the directionality of transport governed by the asymmetric distribution of Ran-GTP across the nuclear envelope. Related references: PMID:7892216
- KPNA1 also participates in the nuclear import of the PR-DUB complex component BAP1, acting redundantly with KPNA2 and TNPO1. Related references: PMID:35446349
- In the context of infection, KPNA1 can mediate the nuclear import of human cytomegalovirus UL84 protein by recognizing a non-classical NLS.
- Subcellularly, KPNA1 shuttles between the cytoplasm and nucleus, consistent with its transport role.
- It is expressed ubiquitously across human tissues.
- Post-translational modifications including acetylation and phosphorylation have been identified, and the protein is a target for ubiquitination.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-300) of human KPNA1, which contains the importin-beta binding (IBB) domain; consider this when selecting detection reagents or designing competition assays.
- The antibody is validated for WB, IF/ICC, IF-P, IHC-P, and ELISA applications on human, mouse, and rat samples; always validate in your specific sample system and optimize antibody dilutions.
- For IF assays, note that KPNA1 shuttles between cytoplasm and nucleus; permeabilization protocols may require optimization to clearly visualize both compartments.
- As a monoclonal antibody, it offers consistent lot-to-lot performance suitable for quantitative comparative studies.
