Biological Background: IMPA1 Function and Localization
- IMPA1 (inositol monophosphatase 1) catalyzes the dephosphorylation of myo-inositol monophosphates to generate myo-inositol, a critical precursor for phosphatidylinositols and polyphosphoinositides (PubMed:17068342, PubMed:8718889, PubMed:9462881). Related references: PMID:17068342, PMID:8718889, PMID:9462881
- The enzyme also dephosphorylates other sugar-phosphates in vitro, including D-galactose-1-phosphate, glucose-1-phosphate, glucose-6-phosphate, fructose-1-phosphate, beta-glycerophosphate and 2′-AMP (PubMed:17068342, PubMed:8718889, PubMed:9462881). Related references: PMID:17068342, PMID:8718889, PMID:9462881
- IMPA1 is a pharmacological target for lithium (Li+) in brain, and its inhibition may underlie lithium's therapeutic effect in bipolar disorder (PubMed:17068342). Related references: PMID:17068342
- It is essential for maintaining inositol homeostasis and normal brain function; mutations are associated with intellectual disability (PubMed:26416544, PubMed:8718889). Related references: PMID:26416544, PMID:8718889
- The protein localizes predominantly to the cytoplasm.
- Post-translational modifications include phosphorylation; it requires magnesium ions for activity and is metal-binding.
- Alternative splicing generates multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen consists of the full-length recombinant human IMPA1 protein (amino acids 1–277), suggesting the antibody can detect both native and denatured forms; consider this when optimizing for different applications.
- For Western blot (WB), an expected band at approximately 30 kDa should be observed; validate in the relevant human, mouse, or rat tissue lysates.
- In immunohistochemistry (IHC-P) and immunocytochemistry (IF/ICC), since IMPA1 localizes to the cytoplasm, permeabilization steps are required; antigen retrieval methods may influence staining patterns.
- For ELISA, the polyclonal nature of the antibody may provide robust detection; consider pairing with a specific capture antibody or direct antigen coating.
