Biological Background: ILT5/CD85a Function and Localization
- LILRB3 (also known as ILT5, CD85a, LIR3) is a member of the leukocyte immunoglobulin-like receptor subfamily B.
- Functions as a receptor for class I MHC antigens and contains immunoreceptor tyrosine-based inhibitory motifs (ITIM) in its intracellular domain.
- Upon co-ligation with immune receptors such as FCGR2B and the B-cell receptor, LILRB3 down-regulates antigen-induced B-cell activation by recruiting phosphatases.
- Predominantly expressed on the cell membrane of monocytes and B-cells.
- The protein undergoes alternative splicing and post-translational modifications including glycosylation and phosphorylation.
- The molecular weight is approximately 69 kDa (canonical isoform), with a minor isoform at 71 kDa.
Experimental Guidance and Technical Tips
- This -conjugated antibody is designed for direct flow cytometry detection of human ILT5/CD85a on live cells.
- Consider using appropriate isotype controls and Fc receptor blocking steps to reduce non-specific binding.
- The immunogen corresponds to the extracellular region (aa 24-418), which contains multiple Ig-like domains; thus, the antibody is likely to recognize the native conformation of the receptor.
- Titrate the antibody to determine the optimal concentration for your specific experimental conditions.
- Validate the staining pattern using known ILT5-expressing cell lines or primary monocytes and B-cells.
