Biological Background: ILF3 Function and Localization
- ILF3 is a ubiquitously expressed RNA-binding protein that exists as multiple isoforms (e.g., NF90, NF110) due to alternative splicing.
- It plays an essential role in the biogenesis of circular RNAs (circRNAs) by stabilizing flanking intronic regulatory elements to promote back-splicing (PubMed:28625552). Related references: PMID:28625552
- ILF3 binds to poly-U elements and AU-rich elements (AREs) in the 3'-UTR of target mRNAs, thereby regulating transcript stability and translation (PubMed:14731398). Related references: PMID:14731398
- Upon viral infection, ILF3 accumulates in the cytoplasm and participates in the innate antiviral response by inhibiting viral mRNAs after phosphorylation by PKR (PubMed:21123651, PubMed:34110282). Related references: PMID:21123651 PMID:34110282
- Additionally, ILF3 positively regulates HIV-1 virus production by binding to and stabilizing HIV-1 RNA.
- Subcellularly, ILF3 localizes primarily to the nucleus and nucleolus, but translocates to the cytoplasm under specific conditions such as viral infection.
- As reflected by its keyword annotations, ILF3 is subject to various post-translational modifications including phosphorylation, acetylation, methylation, and ubiquitination, and functions in transcription regulation and antiviral defense.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-100) of ILF3; this sequence may be conserved across known splice variants, allowing detection of multiple isoforms.
- In western blotting (WB), the antibody is expected to detect a primary band at ~95kDa, but additional bands (e.g., ~110kDa for NF110) may be observed due to alternative splicing; validate the pattern using appropriate cell lysates.
- For immunohistochemistry (IHC-P), optimize antigen retrieval conditions and include control tissues (e.g., brain, testis) given the ubiquitous expression of ILF3.
- When used in ELISA, consider including a peptide blocking control to confirm specificity.
