Biological Background: ILF3 Function and Localization
- ILF3, also known as NF90, DRBP76, and MPP4, is a ubiquitously expressed RNA-binding protein with multiple roles in gene expression regulation.
- It is essential for circular RNA (circRNA) biogenesis by stabilizing regulatory elements in flanking introns to promote back-splicing.
Related references: PMID:28625552 - ILF3 binds to poly-U and AU-rich elements (AREs) in the 3'-UTR of target mRNAs, influencing mRNA stability and translation.
Related references: PMID:14731398 - Upon viral infection, ILF3 is phosphorylated by double-stranded RNA-activated protein kinase (PKR), releasing it from circRNAs to interact with and inhibit viral mRNAs, contributing to innate antiviral defense.
Related references: PMID:21123651 PMID:34110282 - In HIV-1 infection, ILF3 promotes virus production by binding and stabilizing HIV-1 RNA.
- Subcellularly, ILF3 predominantly localizes to the nucleus and nucleolus but can shuttle to the cytoplasm, particularly during viral infection.
- The protein is a phosphoprotein subject to acetylation, methylation, and ubiquitination, and it features DNA-binding and double-stranded RNA-binding motifs, underscoring its roles in transcription and post-transcriptional regulation.
Experimental Guidance and Technical Tips
- The immunogen used for this antibody corresponds to the N-terminal region (amino acids 1-270) of human ILF3. For Western blotting, expect a band around 95 kDa; consider using appropriate positive and negative controls.
- This polyclonal antibody has been documented for use in WB, IHC-P, IF/ICC, IP, and ELISA. Due to the ubiquitous expression of ILF3, validate the antibody in your specific sample type and tissue.
- For IHC-P and IF/ICC, optimize antigen retrieval and permeabilization conditions, and include appropriate nuclear counterstains to confirm localization.
