Biological Background: IL9 Function and Localization
- Interleukin-9 (IL9), also known as cytokine P40 or T-cell growth factor P40, is a multifunctional cytokine belonging to the IL-7/IL-9 family. It is primarily secreted by T-helper 2 (Th2) lymphocytes, mast cells, and NKT cells.

- This cytokine promotes the differentiation of IL-17-producing helper T-cells (Th17) and stimulates the proliferation and differentiation of mast cells.
- IL9 signals through a heterodimeric receptor complex consisting of the IL9R subunit and the common gamma chain (IL2RG), leading to activation of JAK1 and JAK3 kinases.
- Downstream signaling involves STAT1, STAT3, and STAT5; STAT1 primarily mediates differentiation gene induction, while STAT3 and STAT5 protect cells from apoptosis.
- The protein is secreted and undergoes post-translational modifications, including glycosylation and formation of pyrrolidone carboxylic acid at the N-terminus.

Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 19-144 of human IL9, representing the mature protein after signal peptide cleavage. This design targets the biologically active secreted form.
- For Western blotting, a band corresponding to ~16 kDa is expected under reducing conditions. Due to glycosylation, higher molecular weight species may be observed; validate in the appropriate sample type.
- In immunohistochemistry (paraffin-embedded sections), ensure antigen retrieval conditions are optimized for the tissue and fixation method used. Consider including relevant positive and negative tissue controls.
- This polyclonal antibody is also suitable for ELISA applications; however, pairing with a monoclonal detection antibody may improve specificity in sandwich ELISA formats. The user should validate the assay in their own system.
