Biological Background: IL6ST Function and Localization
-
IL6ST, also known as gp130, CD130, or interleukin-6 receptor subunit beta, is a ubiquitously expressed transmembrane protein that serves as the common signal-transducing subunit for the IL-6 cytokine family, including IL-6, LIF, OSM, CNTF, IL-11, CTF1, and others. Related references: PMID:11285233, PMID:11294841, PMID:1542794
-
Upon cytokine binding, IL6ST undergoes homodimerization (for IL-6 or IL-11) or heterodimerization with LIFR or OSMR (for other family members), leading to activation of JAK tyrosine kinases and downstream STAT signaling, as well as YAP and NOTCH pathways. Related references: PMID:19915009, PMID:2261637, PMID:23294003, PMID:39532904, PMID:8999038
-
IL6ST also binds the soluble IL-6/sIL-6R complex (hyper-IL-6), inhibiting IL-6 trans-signaling and the acute phase response, and it can block IL-11 cluster signaling through IL-11R. Related references: PMID:11121117, PMID:21990364, PMID:30279168
-
The protein is located on the cell membrane and can also be secreted; it is found in all tissues and cell lines tested and is present in blood serum. Related references: PMID:2261637, PMID:24629561
-
IL6ST is a glycoprotein with a calculated molecular weight of ~103.5 kDa (observed at ~104 kDa), featuring immunoglobulin-like domains, disulfide bonds, and multiple phosphorylation sites; it plays roles in immune response, hematopoiesis, bone metabolism, pain control, and embryonic development.
-
The gene for IL6ST (Gene ID 3572 in human) gives rise to multiple isoforms via alternative splicing; mutations are associated with several diseases including hyper-IgE recurrent infection syndrome and inflammatory bowel disease.
Experimental Guidance and Technical Tips
- The immunogen sequence (aa 678–857) lies within the intracellular region, so the antibody may recognize both full-length and truncated forms; verify specificity in your experimental system.
- For Western blotting, expect a band at ~104 kDa; additional bands may represent splice isoforms or degradation products—optimize sample preparation and loading controls.
- This polyclonal antibody has been reported to work in IHC-P and ELISA; for IHC-P, consider antigen retrieval methods and test recommended dilutions starting from 1:50 to 1:200.
- Cross-reactivity with mouse and rat is predicted based on sequence homology; validate in the relevant sample system if working with these species.
