Biological Background: IL6ST Function and Localization

  • Interleukin-6 receptor subunit beta (IL6ST, also known as gp130, CD130, CDw130, or oncostatin-M receptor subunit alpha) is a single-pass type I membrane protein that serves as a common signal-transducing receptor for the IL-6 cytokine family.
  • IL6ST functions as the shared signal-transducing subunit for IL-6, IL-11, LIF, OSM, CNTF, CTF1, CLCF1, and other cytokines. Ligand binding induces receptor homodimerization (for IL-6/IL-11) or heterodimerization with LIFR or OSMR, initiating intracellular signaling cascades.   • Receptor dimerization activates associated JAK kinases, which phosphorylate the intracellular domains, recruiting and activating STAT transcription factors (primarily STAT3) to regulate expression of target genes.   • This polyclonal antibody was raised against the intracellular domain (aa 751-918) of human IL6ST; for Western blotting, expect a band at approximately 104 kDa (may appear higher due to glycosylation). Validate under reducing conditions.
  • For immunofluorescence/ICC, intracellular staining may require cell permeabilization. Optimize fixation and permeabilization protocols for the target cell type.
  • Reported cross-reactivity with mouse and rat IL6ST suggests utility across these species, but it is recommended to verify reactivity in the relevant sample system.
