Biological Background: IL6R Function and Localization

• IL6R (interleukin-6 receptor subunit alpha) is a single-pass type I membrane glycoprotein belonging to the immunoglobulin superfamily. It is also designated gp80, CD126, and IL-6R alpha chain.
• Membrane-bound IL6R serves as the ligand-binding alpha subunit for interleukin-6 (IL6). The IL6/IL6R complex associates with the signal transducing subunit IL6ST/gp130 to initiate classic signaling, which is primarily regenerative and anti-inflammatory and is restricted to tissues such as the liver and certain immune cells.
• The soluble form of IL6R (sIL6R), generated by alternative splicing and ectodomain shedding, acts as an agonist of IL6 activity. The IL6:sIL6R complex (hyper-IL6) binds to IL6ST/gp130 on cells lacking membrane IL6R, enabling 'trans-signaling' that is pro-inflammatory and crucial in chronic inflammatory diseases (PubMed:21990364). This complex also induces VEGF production (PubMed:12794819). In plasma, 1–20% of total sIL6R is generated by alternative splicing (PubMed:28060820). • IL6R signaling also drives the differentiation of naïve CD4+ T cells to the Th17 lineage through 'cluster signaling' by dendritic cells.

• In immunohistochemistry (IHC-P), this antibody detects membrane-localized IL6R. Consider antigen retrieval optimization and include appropriate controls to confirm specificity.
• For ELISA applications, this polyclonal antibody can be used for capture or detection, but note that the presence of IL6 or soluble IL6R may affect assay design; cross-validation with recombinant standards is recommended.
