Biological Background: IL4R Function and Localization

Interleukin-4 receptor subunit alpha (IL4R, also known as CD124 or IL4RA) is the high-affinity receptor subunit for both interleukin 4 (IL4) and interleukin 13 (IL13). Upon ligand binding, it couples to the JAK1/2/3-STAT6 signaling pathway, driving Th2 differentiation and regulating IgE production, chemokine and mucus secretion in allergic inflammation. A soluble isoform (sIL4R) is produced by alternative splicing or proteolytic cleavage; it acts as a decoy receptor that inhibits IL4-mediated cell proliferation and IL5 up-regulation by T-cells.
