Biological Background: IL4I1 Function and Localization
- Protein identity and isoforms: IL4I1 (L-amino-acid oxidase, also known as interleukin-4-induced protein 1 or FIG1) is a secreted flavoprotein belonging to the L-amino acid oxidase family. Encoded by the IL4I1 gene, it generates multiple isoforms through alternative splicing and alternative promoter usage.
- Core enzymatic function: IL4I1 catalyzes the oxidative deamination of L-amino acids, with preference for aromatic residues: phenylalanine, tyrosine, and tryptophan are converted to phenylpyruvic acid, hydroxyphenylpyruvic acid, and indole-3-pyruvic acid, respectively. - Immunoregulatory role via AHR activation: By degrading tryptophan, IL4I1 generates indole metabolites (indole-3-pyruvic acid, indole-3-acetic acid, indole-3-aldehyde) and kynurenic acid that act as ligands for the aryl hydrocarbon receptor (AHR). - Modulation of immune cell function: IL4I1 inhibits T-cell activation and proliferation, promotes the differentiation of naive CD4+ T cells into FOXP3+ regulatory T cells, and regulates B-cell development. - Subcellular localization: IL4I1 is secreted and additionally found in lysosomes, cytoplasmic vesicles, and secretory vesicles including acrosomes.
- Tissue and disease expression: Highest expression occurs in immune tissues (lymph nodes, spleen), with protein detected in germinal center macrophages, inflammatory myeloid cells, antigen-presenting cells, and spermatozoa. It is highly expressed in primary mediastinal large B-cell lymphoma and other B-cell lymphomas, as well as in tumor-associated macrophages.
