Biological Background: Interleukin-3 receptor subunit alpha Function and Localization
- The IL3RA gene encodes the interleukin-3 receptor alpha subunit (CD123), a type I transmembrane protein belonging to the cytokine receptor family.
- IL3RA is primarily expressed on hematopoietic progenitor cells, monocytes, and B-lymphocytes, where it controls the proliferation and differentiation of these cells into committed lineages. Related references: PMID:10527461
- Upon IL-3 ligand binding, IL3RA associates with the common beta chain (IL3RB) to form a high-affinity receptor complex, triggering intracellular signal transduction.
- Ligand stimulation is reported to activate JAK2 and PI3K, which in turn phosphorylate and activate STAT5, driving transcriptional programs that promote cell survival and proliferation.
- IL3RA is located on the cell membrane and undergoes post-translational modifications including disulfide bond formation, glycosylation, and ubiquitination.
- Alternative splicing generates multiple IL3RA isoforms, and the protein has been structurally characterized by X-ray crystallography.
Experimental Guidance and Technical Tips
- For Western blotting, the predicted molecular weight is approximately 43 kDa; validate signal specificity using IL3RA knockout or knockdown controls.
- In IHC-P applications, consider heat-induced antigen retrieval and test primary antibody dilution series to optimize staining; include appropriate isotype controls.
- For ELISA, the antibody can be used as a capture or detection reagent; pair with a matched monoclonal antibody to develop a sandwich ELISA system.
- Since the immunogen lies within the N-terminal extracellular domain (aa 1-100), the antibody may detect multiple isoforms if they retain this region; cross-reactivity with mouse and rat IL3RA has been reported by the supplier, but validate in your experimental system.
