Biological Background: IL36R Function and Localization
- Interleukin-1 receptor-like 2 (IL36R), also known as IL-1Rrp2 or IL-1 receptor-related protein 2, is a receptor for the interleukin-36 cytokines (IL36A, IL36B, and IL36G) and belongs to the interleukin-1 receptor family.
- Upon ligand binding, IL36R associates with the coreceptor IL1RAP to form a functional receptor complex, which triggers downstream signaling cascades including NF-kappa-B and MAPK pathways (By similarity).
- The IL-36 signaling system is localized to epithelial barriers and plays a role in local inflammatory responses, analogous to the IL-1 system, and is particularly implicated in skin inflammation through the induction of the IL-23/IL-17/IL-22 pathway.
- IL36R is a membrane protein with a calculated molecular weight of approximately 65.4 kDa, and it undergoes post-translational modifications including glycosylation and disulfide bond formation.
- Tissue expression of IL36R is observed in synovial fibroblasts, articular chondrocytes, keratinocytes, monocyte-derived dendritic cells, monocytes, and myeloid dendritic cells, as confirmed at the protein level.
- The receptor features extracellular immunoglobulin-like domains and a cytoplasmic TIR domain, characteristic of the interleukin-1 receptor family.
- Alternative splicing of the IL1RL2 gene leads to multiple transcript variants.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal extracellular domain (amino acids 1-116), which may recognize both full-length and extracellular domains of IL36R. Consider this when designing detection assays for soluble receptor forms or membrane-bound receptor.
- For Western blot (WB), the predicted molecular weight is 65 kDa; however, glycosylation may cause the band to appear higher. Validate in relevant cell lysates known to express IL36R (e.g., keratinocytes, monocytes).
- For immunofluorescence/immunocytochemistry (IF/ICC), membrane localization is expected. Include appropriate permeabilization controls to distinguish surface staining from intracellular pools.
- Cross-reactivity with mouse IL36R is reported, making this antibody suitable for studies in mouse models. However, always validate specificity in your experimental system.
