Biological Background: IL36A Function and Localization
- Interleukin-36 alpha (IL36A), also known as FIL1 epsilon, IL-1 epsilon, or IL-1F6, is a cytokine member of the IL-1 family.
- Part of the IL-36 signaling system, it binds to the IL1RL2/IL-36R receptor, activating NF-kappa-B and MAPK pathways to promote pro-inflammatory responses.
- Acts on keratinocytes, dendritic cells, and T-cells in epithelial barriers, driving tissue infiltration, cell maturation, and proliferation; induces chemokines (CCL3, CCL4, CCL5, CXCL8, etc.) and cytokines (TNF, IL-6, IL-8).
- Predominantly expressed in skin keratinocytes and upregulated in lesional psoriasis skin, playing a key role in inflammatory skin diseases.
- In cultured monocytes, up-regulates IL-1A, IL-1B, and IL-6; in dendritic cells, promotes maturation and T-cell proliferation.
- Found in the cytoplasm and secreted; tissue expression includes immune system and fetal brain, but highly specific to skin keratinocytes (not detected in fibroblasts, endothelial cells, or melanocytes).
- Keywords: 3D-structure, cytokine, innate immunity, inflammatory response, nitration, secreted, proteomics identification.
- May also play a role in pro-inflammatory effects in the lung.
Experimental Guidance and Technical Tips
- The immunogen covers full-length human IL36A (aa 1-158); thus, the antibody likely recognizes both precursor and mature forms. For WB, expect a band around 18 kDa.
- When performing WB, include positive and negative controls (e.g., recombinant protein or cell lysates from skin keratinocytes) to confirm specificity.
- For ELISA applications, optimize coating conditions and antibody dilutions using recombinant IL36A standard curves.
- This antibody demonstrates cross-reactivity with mouse and rat IL36A, enabling its use in rodent models of inflammatory skin diseases; nevertheless, verify species-specific reactivity empirically.
- Due to the cytokine's secretion, consider analyzing both cell lysates and culture supernatants in functional studies.
- The antibody is validated for WB and ELISA only; other applications (IHC, IF) may require additional optimization and validation.
