Biological Background: IL-33 Function and Localization
- IL-33 (also known as Il-33, Il1f11, NF-HEV, or 9230117N10Rik) is a dual-function protein of the IL-1 cytokine family. The full-length precursor can be processed into mature cytokine forms.
- As a cytokine, IL-33 binds to the IL1RL1/ST2 receptor and activates NF-κB and MAPK signaling pathways, driving Th2 cell maturation and type 2 cytokine secretion. Related references: PMID:29045903
- IL-33 functions as an alarmin, released upon tissue damage to act as a chemoattractant for Th2 cells and to activate mast cells, basophils, eosinophils, and natural killer cells (By similarity).
- IL-33 induces rapid UCP2‑dependent mitochondrial rewiring in macrophages, lowering reactive oxygen species and sustaining itaconate production to promote GATA3‑dependent differentiation of alternatively activated macrophages. Related references: PMID:34644537
- In quiescent endothelial cells, uncleaved IL-33 resides in the nucleus, associating with chromatin and acting as a transcriptional repressor by sequestering NF-κB/RELA (By similarity).
- Subcellularly, IL-33 is detected in the nucleus, chromosomes, cytoplasm, cytoplasmic vesicles, and can be secreted, reflecting its multifaceted functions.
- The mature protein has a calculated molecular weight of approximately 30 kDa, consistent with the full‑length murine IL-33.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 109‑266 of mouse IL‑33) covers a region within the cytokine domain; the antibody may recognize both full‑length and proteolytically processed forms. Consider this when interpreting bands differing from the predicted ~30 kDa in WB.
- For Western blotting, a band near 30 kDa is expected in mouse and rat tissue or cell lysates. Validate in the relevant sample system and compare with known expression patterns.
- IL‑33 can be detected by ELISA in various biological fluids; for sandwich ELISA, pair this pAb with a validated capture antibody (monoclonal) and perform titration to optimize signal.
- Cross‑reactivity with rat is reported; for other species or specific applications, confirm by pilot experiments.
- Shipping on ice bags ensures antibody stability; upon receipt, aliquot and store at recommended temperatures to maintain activity.
