Biological Background: Interleukin-33 Function and Localization
- IL33 (Interleukin-33), also known as DVS27, IL1F11, and NF-HEV, is a member of the IL-1 cytokine family.
- It signals through the IL1RL1/ST2 receptor, activating NF-kappa-B and MAPK signaling pathways in target cells. Related references: PMID:16286016, PMID:19841166
- IL33 is crucial for Th2 cell maturation, inducing secretion of T-helper type 2-associated cytokines, and activates mast cells, basophils, eosinophils, and natural killer cells. Related references: PMID:17853410, PMID:18836528
- As an alarmin, IL33 acts as a chemoattractant for Th2 cells and amplifies immune responses during tissue injury. Related references: PMID:17853410, PMID:18836528
- It enhances polarization of alternatively activated macrophages and may induce UCP2-dependent mitochondrial rewiring to attenuate reactive oxygen species and preserve Krebs cycle integrity. Related references: PMID:19841166
- In quiescent endothelial cells, uncleaved IL33 acts as a chromatin-associated nuclear factor with transcriptional repressor properties, potentially sequestering NF-kappaB/RELA to reduce target expression; this form is rapidly lost upon angiogenic or pro-inflammatory activation. Related references: PMID:21734074, PMID:18787100
- Subcellularly, IL33 localizes to the nucleus, chromosome, cytoplasm, cytoplasmic vesicles, and can be secreted.
- It is highly expressed in high endothelial venules of tonsils, Peyer's patches, and mesenteric lymph nodes, but nearly absent in placenta.
Experimental Guidance and Technical Tips
- For western blotting, a band near 31 kDa is expected; validate expression in appropriate cell or tissue lysates, considering that IL33 may exist as both full-length and cleaved forms.
- Cross-reactivity with mouse and rat samples is reported; when analyzing rodent tissues, confirm that the antibody recognizes the expected isoform and molecular weight.
- In ELISA applications, the polyclonal antibody may serve as a capture or detection reagent; test optimal pairing with other anti-IL33 clones for sandwich ELISA development.
- The immunogen spans amino acids 110–190 of human IL33, a region that includes residues potentially involved in receptor binding; epitope specificity may be useful in cleavage-site-specific studies.
- Given the nuclear and cytoplasmic localizations of IL33, consider subcellular fractionation or immunostaining protocols to distinguish between chromatin-associated and secreted pools.
