Biological Background: Interleukin-33 Function and Localization
- Interleukin-33 (IL-33), also known as IL-1F11 and NF-HEV, is a dual-function protein encoded by the IL33 gene (C9orf26, IL1F11, NFHEV). It exists as both a secreted cytokine and a nuclear factor, with alternative splicing generating multiple isoforms.
- As a cytokine, IL-33 binds to the IL1RL1/ST2 receptor, activating NF-kappa-B and MAPK pathways. It promotes Th2 cell maturation and secretion of type 2 cytokines, and activates mast cells, basophils, eosinophils, and natural killer cells. Related references: PMID:16286016 PMID:19841166 PMID:17853410 PMID:18836528
- IL-33 enhances alternative macrophage polarization and acts as a chemoattractant for Th2 cells, functioning as an 'alarmin' released upon tissue injury to amplify immune responses. Related references: PMID:19841166 PMID:17853410 PMID:18836528
- The uncleaved nuclear form of IL-33 is highly expressed in quiescent endothelial cells, where it acts as a chromatin-associated transcriptional repressor by sequestering NF-kappaB/RELA, reducing pro-inflammatory gene expression. This form is rapidly lost upon angiogenic or pro-inflammatory activation. Related references: PMID:21734074 PMID:18787100
- IL-33 induces rapid UCP2-dependent mitochondrial rewiring in macrophages, attenuating reactive oxygen species and preserving Krebs cycle integrity for persistent itaconate production, which supports the differentiation of inflammation-resolving alternatively activated macrophages (by similarity).
- Subcellular localization varies between the nuclear form (nucleus, chromosome) and the secreted form (cytoplasm, cytoplasmic vesicles, secretion), reflecting its dual functionality.
- Tissue specificity: Expressed at high levels in high endothelial venules of tonsils, Peyer's patches, and mesenteric lymph nodes; almost undetectable in placenta.

Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 109-270 of human IL33 (NP_001300974.1), which covers the mature cytokine domain. For WB, consider using positive controls such as stimulated endothelial cells or recombinant IL-33, and note that full-length and cleaved forms may both be detected at masses around 31 kDa or lower.
- For IF/ICC and IHC-P, validate nuclear and cytoplasmic staining patterns in relevant tissue sections or cell lines expressing IL-33; perform appropriate controls to confirm specificity.
- ELISA applications can be developed using this antibody as a capture or detection reagent; titrate both antibody and antigen to optimize signal-to-noise ratio.
- Given the dual nuclear and secreted functions, select sample preparation protocols that preserve both compartments if investigating total IL-33 expression.
