Biological Background: IL-26 Function and Localization
- IL-26 (Interleukin-26), also designated AK155, is a secreted cytokine of the IL-10 family.
- It contributes to mucosal immunity and exhibits pro-inflammatory activity, notably in the context of inflammatory bowel disease (IBD).
- IL-26 triggers intracellular signaling through activation of STAT1, STAT3, MAPK1/3 (ERK1/2), JUN, and AKT pathways.
- It induces expression of SOCS3, TNF, IL-8, and surface ICAM1, and stimulates secretion of IL-8 and IL-10.
- IL-26 reduces proliferation of intestinal epithelial cells, and its function is antagonized by heparin.
- The protein is primarily expressed in HVS-transformed T-cells, colonic T-cells (including Th17 cells), and is elevated in serum of Crohn’s disease patients.
- IL-26 contains a signal peptide and is secreted as a mature ~20 kDa protein (UniProt: 19843 Da).
