Biological Background: Interleukin-25 Function and Localization
- Interleukin-25 (IL-25, also known as IL-17E) is a cytokine produced by eosinophils, T-helper type 2 (Th2) cells, and epithelial cells, playing a key role in adaptive immune responses by regulating cytokine production. Related references: PMID:15860795, PMID:25821217
- Promotes and augments Th2 responses both locally and systemically, contributing to type 2 immunity. Related references: PMID:25821217
- Signals through a heterodimeric receptor complex of IL17RA and IL17RB, activating the JAK2-STAT5A pathway and stimulating secretion of type-2 associated cytokines such as IL4, IL9, and IL13. Related references: PMID:25821217
- Induces release of IL8 and IL6 from eosinophils via combined activation of MAPK and NF-kappa-B pathways. Related references: PMID:15860795
- Inhibits differentiation of T-helper 17 (Th17) cells through production of IL4, IL5, and IL13, highlighting its role in balancing Th17/Th2 responses. Related references: PMID:11754819
- Localized to the secreted compartment, consistent with its function as a circulating cytokine.
- Expressed at low levels in various tissues including brain, kidney, lung, prostate, testis, spinal cord, adrenal gland, and trachea.
- Contains post-translational modifications such as a signal peptide, disulfide bonds, and glycosylation; predicted molecular weight of approximately 20 kDa.
Experimental Guidance and Technical Tips
- The immunogen comprises amino acids 33–177 of human IL25 (mature protein region), so the antibody is expected to recognize the secreted, biologically active form.
- For Western blotting (WB), expect a band around 20 kDa under reducing conditions; consider using positive controls such as recombinant IL25 or stimulated eosinophil/Th2 cell lysates.
- For immunofluorescence (IF-P), optimize permeabilization conditions since the target is secreted; co-staining with organelle markers may be helpful.
- For ELISA, the polyclonal nature enables capture/detection pairing; validate performance with relevant standard curves and sample matrices.
- Cross-reactivity with mouse and rat has been reported, making this antibody suitable for translational studies; however, validate in the appropriate species model.
- As a polyclonal antibody, lot-to-lot consistency should be verified; establish internal reference standards for key applications.
