Biological Background: IL22RA1 Function and Localization
- IL22RA1 (interleukin-22 receptor subunit alpha-1) is a component of the heterodimeric receptors for interleukin-20 (IL20), IL22, and IL24. It partners with IL10RB to form the functional IL22 receptor, and with IL20RB to form one of the IL20 and IL24 receptor complexes.
- Upon ligand binding, the IL22RA1-containing complexes activate the JAK/STAT signaling pathways. IL22 also triggers MAPK1/MAPK3 (ERK1/2) and Akt kinase cascades, linking to cellular proliferation and survival.
- IL22RA1 belongs to the class II cytokine receptor family and is also known as CRF2-9 (cytokine receptor family 2 member 9), ZcytoR11, and IL22R.
- The receptor is located on the cell membrane, consistent with its role in extracellular cytokine sensing and signal transduction.
- Post-translational modifications include glycosylation, disulfide bond formation, phosphorylation, and ubiquitination, which may regulate receptor stability and signaling.
- IL22RA1 is expressed in colon, liver, lung, pancreas, and kidney, but is absent in immune cells (monocytes, T-cells, NK-cells). It is found in keratinocytes of normal and psoriatic skin, and in blood-brain barrier endothelial cells, with increased expression in multiple sclerosis lesions.
- Overexpression has been detected in synovial fluid cells from rheumatoid arthritis and spondyloarthropathy patients, suggesting a role in inflammatory diseases.
- IL24, through the IL22RA1/IL20RB receptor, mediates anti-angiogenic activity and inhibits endothelial cell tube formation and differentiation.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 16–230 of human IL22RA1, which corresponds to the extracellular domain; the antibody may recognize the native receptor on cell surfaces in flow cytometry or immunohistochemistry, though these applications are not pre-validated.
- For Western blot, a band near the predicted 63 kDa is expected, but glycosylation and other post-translational modifications may result in a higher apparent molecular weight. Consider using deglycosylation enzymes for precise MW confirmation.
- This polyclonal antibody is raised in rabbit and shows reported cross-reactivity with mouse and rat, making it potentially useful for studies in rodent models. Validate in your specific sample system using appropriate controls.
- ELISA applicability suggests the antibody can capture or detect soluble or immobilized IL22RA1; optimal dilution and buffer conditions should be experimentally determined.
