Biological Background: IL-22 Function and Localization
- Interleukin-22 (IL-22), also known as cytokine Zcyto18 and IL-10-related T-cell-derived-inducible factor (IL-TIF), is a member of the IL-10 cytokine family.
- It plays a critical role in modulating tissue responses during inflammation, particularly in promoting epithelial cell regeneration and barrier function after injury (PubMed:17204547). Related references: PMID:17204547
- IL-22 signals through a heterodimeric receptor composed of IL22RA1 and IL10RB, activating JAK1/TYK2 and STAT3, as well as ERK1/2 and PI3K/AKT pathways (PubMed:10875937, PubMed:18599299, PubMed:25793261, PubMed:31311100). Experimental Guidance and Technical Tips
- The immunogen used was a recombinant protein spanning amino acids 34-179 of human IL22 (NP_065386.1), covering the mature secreted form. This polyclonal antibody may recognize both full-length and cleaved forms of IL-22.
- For western blot (WB), the predicted molecular weight is around 20 kDa. Consider including positive controls such as stimulated immune cell supernatants or recombinant IL22 protein to verify specificity.
- For immunofluorescence (IF/ICC), optimize fixation/permeabilization conditions, as IL-22 is secreted; using protein transport inhibitors may enhance intracellular staining.
- For ELISA, this antibody can be used as a capture or detection antibody; pair with a monoclonal antibody against a different epitope for sandwich ELISA, and validate in the relevant sample system.
