Biological Background: IL-2 Function and Localization

  • IL-2 is a cytokine primarily produced by activated CD4-positive helper T-cells, and to a lesser extent by CD8-positive T-cells and natural killer (NK) cells, playing pivotal roles in immune response and tolerance. Related references: PMID:6438535
  • It binds to a receptor complex composed of either the high-affinity trimeric IL-2R (IL2RA/CD25, IL2RB/CD122, and IL2RG/CD132) or the low-affinity dimeric IL-2R (IL2RB and IL2RG). Related references: PMID:16293754 PMID:16477002
  • Receptor binding triggers oligomerization and conformational changes, leading to JAK1/JAK3 phosphorylation and subsequent STAT5 phosphorylation, activating STAT, PI3K, and MAPK pathways. Related references: PMID:7973659 PMID:8580378 PMID:25142963
  • It functions as a T-cell growth factor and can increase NK-cell cytolytic activity. Related references: PMID:6608729
  • IL-2 promotes strong proliferation of activated B-cells and subsequent immunoglobulin production. Related references: PMID:6438535
  • It is essential for the survival and proliferation of regulatory T-cells, which maintain immune tolerance, and participates in the differentiation and homeostasis of effector T-cell subsets, including Th1, Th2, Th17, and memory CD8-positive T-cells.
  • The protein is secreted and undergoes glycosylation, with a mature form containing a critical disulfide bond.

Experimental Guidance and Technical Tips

  • The immunogen used spans amino acids 21-153, covering the mature secreted form of IL-2. Validate reactivity in your sample system, as recognition may be influenced by post-translational modifications.
  • For SDS-PAGE and Western blot, the predicted molecular weight is 18 kDa; however, glycosylation may cause the observed band to migrate slightly higher, around 15–20 kDa.
  • For IHC-P, antigen retrieval may be required, and lymphoid tissues (e.g., lymph node, spleen) are ideal for initial optimization due to high IL-2 expression.
  • ELISA performance should be confirmed using recombinant IL-2 protein or relevant biological samples. Consider potential interference from soluble receptor fragments or binding proteins.
