Biological Background: IL-2 Function and Localization
- IL-2 is a cytokine primarily produced by activated CD4-positive helper T-cells, and to a lesser extent by CD8-positive T-cells and natural killer (NK) cells, playing pivotal roles in immune response and tolerance. Related references: PMID:6438535

Experimental Guidance and Technical Tips
- The immunogen used spans amino acids 21-153, covering the mature secreted form of IL-2. Validate reactivity in your sample system, as recognition may be influenced by post-translational modifications.
- For SDS-PAGE and Western blot, the predicted molecular weight is 18 kDa; however, glycosylation may cause the observed band to migrate slightly higher, around 15–20 kDa.
- For IHC-P, antigen retrieval may be required, and lymphoid tissues (e.g., lymph node, spleen) are ideal for initial optimization due to high IL-2 expression.
- ELISA performance should be confirmed using recombinant IL-2 protein or relevant biological samples. Consider potential interference from soluble receptor fragments or binding proteins.
